川芎嗪对大鼠血管平滑肌细胞增殖及FGFR1与p21ras表达的干预作用 被引量:2

摘要 目的观察川芎嗪对血管平滑肌细胞(VSMC)增殖及FGFR1、p21ras蛋白表达的影响。方法体外培养大鼠VSMC,建立VSMC增殖模型,应用不同浓度的川芎嗪干预,MTT法测定细胞增殖活力;免疫印迹法检测FGFR1与p21ras蛋白的表达。结果与对照组比较,川芎嗪中、高剂量组细胞增殖活力均显著降低(P<0.05),低剂量组FGFR1与p21ras表达无显著差异(P>0.05),中剂量组、高剂量组的FGFR1与p21ras表达均较明显降低(P<0.05),中、高剂量组表达较低剂量组比显著降低(P<0.05),高剂量组表达又较中剂量组显著降低(P<0.05),呈剂量依赖性。结论川芎嗪能抑制VSMC增殖,该作用与其抑制FGFR1及其相应信号通路中p21ras蛋白的表达有关。 Objective To investigate the effect of Tetramethylpyrazine (TMP) on the proliferation and the expression of FGFR1 and p21ras in VSMCs. Methods Cultured VSMCs proliferation model was established as control, the cultured VSMC treated with TMP in different dose. Cell proliferation activity was detected by MTF method. The expression of FGFR1 and p21ras was detected by Western blot. Results Treated with TMP in different dose, cell proliferation activities were obviously lower than those in control (P 〈 0.05 ). Compared with the control ,the expression of FGFR1 and p21 ras was significantly reduced in middle doses and high doses groups (P 〈0.05);Compared with middle doses group,the expression of FGFR1 and p21ras was clearly reduced in high doses group group(P 〈 0.05 ). Conclusion TMP might play a role of inhibiting vascular smooth muscle cells proliferation by re- ducing the expression of FGFR1 and p21ras.
