灵龟八法按时开穴灸对衰老模型豚鼠血清SOD活性及相关基因表达的影响 被引量:11

摘要 目的探讨灵龟八法按时开穴艾炷灸对衰老模型豚鼠血清SOD活性及相关基因表达(SODmRNA)的影响。方法 60只成年豚鼠随机分成空白对照组(Ⅰ组)、模型对照组(Ⅱ组)、灵龟八法预防组(Ⅲ组)、神阙预防组(Ⅳ组)、灵龟八法治疗组(Ⅴ组)、神阙治疗组(Ⅵ组),每组10只。除空白组外,均采用腹腔注射D-半乳糖溶液建立衰老豚鼠模型。预防组在进行造模的同时分别进行灵龟八法按时开穴艾炷灸、神阙穴艾炷灸,治疗组在造模成功后分别进行灵龟八法按时开穴艾炷灸、神阙穴艾炷灸治疗。观察各组豚鼠生命质量评分、血清SOD活性、肾组织超氧化物歧化酶相关基因(Cu/ZnSODmRNA及Mn-SODmRNA)相对表达量的差异。结果造模结束后,Ⅰ组未出现衰老症状,预防组(Ⅲ、Ⅳ组)出现少许衰老迹象,且血清SOD活性无明显变化,与Ⅰ组相比差异无统计学意义(P>0.05);Ⅱ组及治疗组(Ⅴ、Ⅵ组)出现明显的衰老症状,血清SOD活性显著降低,与Ⅰ组相比较差异显著(P<0.01)。实验结束后,Ⅱ组的血清SOD活性与造模后无明显改变;两个治疗组血清SOD活性升高,与造模前相比差异无统计学意义(P>0.05),与Ⅰ组相比差异无统计学意义(P>0.05);预防组与治疗组的肾组织Cu/Zn-SODmRNA和Mn-SODmRNA的相对表达量接近Ⅰ组,与Ⅱ组比较差异显著(P<0.01);两个预防组SOD活性及肾组织Cu/Zn-SODmRNA和Mn-SODmRNA的相对表达量组间比较差异无统计学意义(P>0.05);两个治疗组SOD活性及肾组织Cu/Zn-SODmRNA和Mn-SODmRNA的相对表达量组间比较差异有统计学意义(P<0.05),灵龟八法按时开穴艾炷灸对衰老模型豚鼠的治疗作用优于神阙穴艾炷灸。结论灵龟八法延缓衰老机理可能与调节SOD及其相关基因表达密切相关,对衰老模型豚鼠具有明显的防治作用;灵龟八法按时开穴艾炷灸对衰老模型豚鼠的治疗作用优于神阙穴艾炷灸。 Objective To investigate the effects of moxibustion according to opening points on time in eight methods of intelligent turtle on Serum SOD activity and expression of related gene. Methods 60 healthy and adult guinea pigs were randomly divided in- to 6 groups, a blank control group( group Ⅰ ), a control group of aging model( group Ⅱ ), a prevention group with eight method of intelligent turtle (group m ), a prevention group with moxibustion at Shenque (RN8) (group Ⅳ ), a group of treatment group with eight methods of intelligent turtle( group Ⅴ ) and a treatment group with moxibustion at Shenque (RN8) (groupⅥ). There were 10 guinea pigs in every group. All groups used intraperitoneal injection of D - half lactose to make aging model except the blank control group. The prevention groups received the treatments both moxibustion according to opening points on time in eight methods of intelligent turtle and moxibustion at Shenque ( RN8 ) during making model. The treatment groups received the treatments both moxibustion according to opening points on time in eight methods of intelligent turtle and moxibustion at Shenque ( RN8 ) after fin- ished making model. To observe groups of guinea pigs'life quality score, serum SOD activity, renal tissue superoxide dismutase genes ( Cu' Zn - SODmRNA and Mn - SODmRNA ) relative expression difference. Results After modeling, the guinea pigs from group Ⅰ showed no symptoms of aging, as for the ones from moxibustion - while - modeling groups ( group Ⅲ and IV ), only mild symptoms of aging were observed,and the serum SOD activity showed no significant change,comparing with group I there was no statistical meaning between them (P 〉 0.05) ; the group Ⅱ and the treatment groups showed sign of aging, serum SOD activity was significantly decreased ; compared with the group I, there were significant differences ( P 〈 0.01 ). After the experiment, for the serum SOD activity in group Ⅱ , there was no obvious change before and after modeling ,two treatment group serum SOD activity increased, compared with that before modeling, there was no statistically significant(P 〉 0.05 ) ;compared with the group I, there was no statistically significant( P 〉 0.05 ), the prevention groups and treatment groups'renal tissue Cu/Zn- SODmRNA and the relative expression of Mn -SODmRNA was close to the group Ⅰ, compared with the group Ⅱ , there were significantly different (P 〈 0.01 ). However,two prevention groups'SOD activity and Cu/Zn - SODmRNA in renal tissue and the relative expression of Mn - SODmRNA the difference between groups was no statistically significant ( P 〉 0.05 ) ; two treatment groups'activity of SOD and Cu/Zn - SODmRNA in renal tissue and the relative expression of Mn - SODmRNA groups had significant difference ( P 〈 0.05 ). The efficacy of regular moxibustion with eightfold method of the sacred tortoise on aging models of guinea pigs was more pro- nounced than those receiving the moxibustion on Shenque ( RN8 ). Conclusion The mechanism of defer - aging with the eight methods of intelligent turtle may be closely related to the SOl) regulation and the expression of related gene. Moxibustion according to opening points on time in eight methods of intelligent turtle has obvious prevention and treatment effect on aging model guinea pigs, and it has a better therapeutic effect than moxibustion at Shenque (RN8).
