水稻叶片中脉发育调控基因OsEL的克隆和功能研究

Cloning and Characterization of Gene OsEL Related to the Midrib Development of Leaf in Rice

摘要 通过Affymetrix水稻表达芯片数据筛选出1个受低温和高温诱导高水平表达的基因OsEL(Erect Leaf,EL),该基因的蛋白序列含有SUI1保守结构域,可能为翻译起始因子蛋白。克隆OsEL基因并构建植物过表达载体,通过农杆菌介导转化水稻台北309,发现该基因的过量表达引起水稻株型发生变化,转基因植株的叶片变直立,叶片中脉变粗,中脉中的气腔和维管束数目增加,分蘖数增加。推测OsEL基因可能通过调控维管束细胞增殖影响叶脉的发育。 Rice ( Oryza sativa L. ) is one of the most important food crops in the world. Rice plant architecture is closely related to rice breeding and it also affects rice grain yield. Under cold or heat stress, gene OsEL( erect leaf, EL) was induced to a higher level, as revealed by microarray analysis using the GeneChip Rice Genome Array ( Af- fymetrix). The sequence analysis showed that the protein encoded by OsEL contained the SUI1 domain which is the conservative domain of the translation initiation factor SUI1 family. Gene OsEL was cloned and sequenced. Its over- expression vector pCAMBIA1300-163-OsEL was constructed and transformed into rice variety Taibei 309 through the Agrobacterium-mediated method. The transgenic plants over-expressing OsEL showed erect leaves with thicker midribs than the wild type plants. The midribs of transgenic plants developed more air cavities and vasculars than those of the control. Gene OsEL is probably involved in the midrib development in rice by regulating cell proliferation of vascular.
