摘要
采用酵母菌对苯酚进行了降解实验,利用紫外—可见吸收光谱扫描实验确定紫外检测器检测波长,建立以苦味酸为内标高效液相色谱(HPLC)对苯酚以及中间产物定量进行测定的检测体系,得到苯酚、邻苯二酚的标准曲线,两者线性相关系数均大于0.99;通过回收实验得到苯酚、邻苯二酚的回收率分别为99.25%、99.43%。通过酵母菌对500mg/L模拟苯酚废水进行降解实验,在转速150r/min、温度30℃时,经过84h降解,苯酚降解率为90.2%,具有良好的降解作用。
Experiment of phenol biodegradation was conducted using yeast as degrading bacteria. High perform-ance liquid chromatography (HPLC) was applied for quantitative analysis of phenol and intermediate products pro-duced during degradation process. In the detection system,picric acid was selected as internal standard of HPLC,and ultra-visible spectrophotometer scanning experiments were performed to determine the wavelength of UV detector. The standard curve of phenol and catechol was obtained,the correlation coefficients of two standard curves were grea-ter than 0.99. The recovery test showed that the recovery rate of phenol and catechol was 99.25% and 99.43% re-spectively. Yeast achieved perfect phenol degradation efficiency when the phenol concentration was 500 mg/L,stirring speed was 150 r/min and degradation temperature was 30 ℃ ,the phenol removal rate reached to 90.2% after 84 h of biodegradation.
