摘要
为了解三穗鸭肌肉生长抑制素(MSTN)基因SNPs与屠宰性状的相关性,本研究采用PCR产物直接测序的方法对三穗鸭肌肉生长抑制素基因外显子的多态性进行检测,结果在研究群体中发现6个SNP位点:SNP1(G76A)、SNP2(A90G)、SNP3(G129A)、SNP4(G234A)、SNP5(A255C)、SNP6(G276A)。采用固定效应模型分析基因多态性与屠宰性状的相关关系,结果显示:在SNP1位点,BB基因型个体的腿肌重、腿肌率和瘦肉率平均值显著高于AA和AB基因型;在SNP2位点,CD基因型个体的胸肌率平均值显著高于CC基因型,SNP4和SNP5在本试验中完全连锁,HH/NN基因型个体的半净膛重和全净膛重平均值显著高于GG/MM和GH/MN基因型,SNP3和SNP6的不同基因型对屠宰性状各指标效应不显著。研究揭示MSTN基因的多态性对三穗鸭屠体性状具有重要影响。
To understand the relationship between single nucleotide polymorphism sites (SNPs)of myostatin (MSTN) gene and slaughter traits in Sansui duck,direct sequencing of PCR products was used to analyze the polymorphisms of the exon of MSTN gene in Sansui duck. The results showed that there were six SNPs in the sample groups,they were SNP1 (G76A), SNP2 (A90G), SNP3 (G 129A), SNP4 ( G234A), SNP5 (A255C) and SNP6 (G276A) respectively. Correlation analysis between SNPs and slaughter traits was carried out by fixed effects model. The results indicated that for SNP1 the duck with BB genotype had higher leg muscle weight,leg muscle percentage and lean meat percentage than AA and AB genotype;for SNP2 CD genotype had higher breast muscle percentage than CC genotype. SNP4 and SNP5 of the third exon was completely linked in this experiment, the duck with HH/NN genotype had higher semi-eviscerated weight and eviscerated weigh than GG/MM and GH/MN duck. There were no significant effect oil slaughter traits in SNP3 and SNP6. This study revealed that the polymorphism of MSTN gene had important influence on slaughter traits.
