摘要
考察新疆塔里木马鹿茸干品多肽的降糖活性。马鹿茸干品经酸提醇沉获得总肽,利用超滤、层析等技术对总肽进行分离纯化。采用高脂饮食加小剂量多次腹腔注射链脲佐菌素(STZ)的方法建立糖尿病小鼠模型,连续腹腔注射总肽5 w,每周测定小鼠空腹血糖。MTT法检测纯化所得马鹿茸多肽对STZ损伤的胰岛瘤细胞RINm5f的修复作用。体内实验表明,总肽高中低剂量组与糖尿病模型组比较,小鼠空腹血糖水平显著降低,且呈剂量依赖性。分离得到相对分子质量为7 214.36的多肽DAP,该多肽能极显著促进损伤后RINm5f的细胞修复。证明马鹿茸干品总肽具有抗糖尿病的作用,多肽DAP能促进损伤后RINm5f的细胞修复,可能是其降糖活性成分之一。
To investigate the hypoglycemic activity of peptides from dry velvet antler of red deer Cervus elaphus Linnaeus, the total peptides were extracted with acetic buffer (pH 3.5 ), deposited in 40% alcohol solution. Peptides were purified by ultrafiltration, gel filtration and reverse phase chromatography. Diabetic model of male mice was established by high-fat diet combined with multiple low-dose streptozotocin (STZ)intraperitoneal injection. Diabetic mice were treated with total peptides from red deer antler for 5 weeks and fasting blood glucose was measured once a week. MTT assay was conducted to determine the effects of the pure peptide isolated from red deer antler on the recovery of injury RINmSf cells by STZ. Compared with the diabetic group, the level of fasting blood glucose was decreased after treatment with antler peptides and showed a dose-dependent manner. A peptide named DAP was purified from dry velvet antler. The molecular weight of DAP was measured to be 7214.36 Dahons by MALDI-TOF MS. After treat- ment with DAP,the viability of RINmSf ceils in the presence of STZ was significantly increased. The total peptides from red deer antler showed an antidiabetic effect and the pure peptide, named DAP, was probably one of the hypoglycemic active components in the dry red deer antler peptides.
