摘要 目的:探讨茶多酚对心室颤动大鼠心肺复苏后存活时间及脑组织活性氧(ROS)的影响。方法:健康SD大鼠随机分为假手术(SH)组、缺血再灌注(IR)组、茶多酚(TP)组、JNK通路阻断剂SP600125(SP)组、TP+SP组。除SH组外均经食道交流电刺激建立心室颤动模型,5 min后行心肺复苏,自主循环恢复(ROSC)即刻随机给药,每组按12、24、48、72 h分为4个时间点。观察ROSC后大鼠72 h内存活时间;检测各时间点脑组织ROS、超氧化物歧化酶(SOD)、丙二醛(MDA)含量。结果:TP组、SP组及TP+SP组大鼠生存时间均较IR组延长(P<0.05)。TP组、SP组及TP+SP组ROS、MDA含量在各时间点均较IR组下降明显(P<0.05)且各组ROS均在24 h、MDA于48 h达高峰。TP组、SP组及TP+SP组脑SOD活性较IR组升高(P<0.05)且均在48 h达最低。结论:TP可延长室颤大鼠复苏后的生存时间,此作用与降低脑ROS、MDA含量,增加SOD活性有关,并可能涉及JNK通路。 Objective To study the effects of tea polyphenols on the survival time and the cerebral tissue reactive oxygen species (ROS) in rats with ventricular fibrillation following cardiopulmonary resuscitation. Methods The Sprague-Dawley rats were randomly divided into the sham (SH), the ischemia reperfusion (IR), the tea polyphenols (TP), the SP600125(SP) and the TP + SP groups. The SD rats were used to establish the ventricular fibrillation (VF) models through transesophageal alternating current stimulation method except those in the SH group. Five minutes after onset of VF, conventional cardiopulmonary resuscitation (CPR) was initiated and intravenous injection was given immediately after restoration of spontaneous circulation (ROSC). The survival time within 72 h after ROSC was observed, brains were collected to determine ROS, superoxide dismutase (SOD) and malonylaldehyde (MDA) at 12 h, 24 b, 48 h and 72 h after VF, respectively. Results Compared to the IR group, the survival times of rats in the treatment groups were significantly prolonged (P 〈 0.05). (2) The level of ROS and MDA in cerebral tissue in the treatment groups were significantly decreased than those in the IR group (P 〈 0.05, respectively), and both of them reached the peak at 24 h and 48h in the other four groups, respectively. (3) The content of SOD in the treatment group was significantly higherthan that in the IR group (P 〈 0.05), and the SOD level in other groups had the minimum value at 48 h after VF. Conclusion TP can prolong the survival time of resuscitation by reducing ROS and MDA level, elevating the SOD activity.
