应用辛酸硫酸胺法提取小鼠腹水和血清中的IgG抗体 被引量:18

摘要 目的:从小鼠腹水和血清中纯化 IgG抗体。方法:首先用辛酸沉淀小鼠腹水和血清中的非免疫球蛋白,然后用固体硫酸胺提取抗人 T和 B细胞白血病单克隆抗体 (IgG1和 IgG2b)和多克隆 IgG抗体。结果:经 SDS-PAGE和活细胞免疫荧光法鉴定,获得了纯度高、活性好的抗体,腹水中单抗的回收率达 80%以上。结论:本方法是一种简单、价廉且有效的纯化小鼠腹水和血清 IgG抗体的方法。 ∶ Objective∶ To purify mouse IgG antibody from the ascites fluid and the serum.Methods∶ In the first step,we used Caprylic acid to precipitate the non-immunoglobulin in the ascites fluids that contain mouse anti-human T and B lymphocytes leukemia(IgG1 and IgG2b)monoclonal antibodies and murine serum.In the second,the IgG fraction was precipitated with Ammonium sulfate.Results∶ Greater than 80% of IgG in mouse ascites fluid and mouse serum could be isolated by this procedure.The antibodies show a high purity and good activities monitored by SDS-PSGE and Live Cell Immunofluorence.Conclusion∶ The procedure is simple,low cost and effective for isolated IgG antibodies from mouse ascites fluid and serum.
