摘要 通过使用可抑制还原型辅酶Ⅱ(NADPH)氧化酶活性、降低细胞内源性活性氧(ROS)的抑制剂二苯基碘(DPI),以及用抗氧化剂N-乙酰半胱氨酸(NAC)2种方法降低单核细胞增生李斯特菌(Listeria monocytogenes,LM)中ROS的产生,同时用外源补充H2O2的办法来验证ROS对LM菌膜(BF)形成的调控作用。结果表明:DPI和NAC能够通过降低ROS的产生进而抑制LM菌膜的生成,细菌中外源补充活性氧(H2O2)对BF的生成有一定的促进作用。此研究说明,和高等动植物一样,LM中也存在类似于负责产生ROS的NADPH氧化酶,由其介导产生的ROS对LM的BF形成不可或缺,ROS可能是BF形成过程中重要的信号分子。 In order to explore the role of ROS (reactive oxygen species) in Listeria monocytogenes (LM) biofilm formation, we added either diphenyleneiodonium chloride (DPI) as a NADPH oxidase inhibitor or N-acetylcysteine (NAC) as an antioxidant in combination with exogenous H2O2 to observe the changes of ROS and biofilm formation. The results showed that both DPI and NAC were able to restrain the biofilm formation of LM by reducing the amount of ROS generation. To some extent, the addition of H2O2 could promote biofilm formation. Our results suggest that NADPH oxidase exists in LM, which is responsible for generating ROS. ROS plays a vital role in LM biofilm formation and may be an important signal molecule.
