硫酸镁对急性肺动脉高压实验室模型的血流动力学影响 被引量:1

EFFECT OF MAGNESIUM-SULFATE ON BLOOD FLOW DYNAMICS IN ACUTE PULMONARY HYPERTENSION MODEL

摘要 目的探讨硫酸镁(MgSO_4)对实验室急性肺动脉高压模型的影响机制。方法 30只杂种犬,随机分为5组,空白组(control)、肺动脉高压(pulmonary hypertension,PH)组以及MgSO_4 7.0、MgSO_4 8.0和MgSO_4 9.0组,其中Control组不给于任何处理,PH组为急性肺动脉高压(acute pulmonary hypertension,APH),其余MgSO_4 7.0、MgSO_4 8.0及MgSO_4 9.0组造成APH稳定30min后,分别静脉给予MgSO_4并维持血MgSO_4浓度为7.0mmol/L、8.0mmol/L及9.0mmol/L。肺动脉、股动脉插管检测平均肺动脉压(mean pulmonary artery pressure,MPAP)、体循环平均动脉压(mean arterial pressure,MAP),静脉取血检测血浆APH血浆内皮素1(endothelin-1,ET-1)、血栓素(thromboxane A_2,TXA_2)以及6-酮前列腺素(6-keto-PGFla)。结果①PH组MPAP较Control组显著升高,MgSO_4 7.0、MgSO_4 8.0及MgSO_4 9.0组MPAP均较PH组显著下降,其中MgSO_4 9.0下降最明显;②PH组MAP较Control组显著降低,其余3组MAP较PH组显著升高,其中MgSO_4 9.0升高最明显。③PH组ET-1、TXA_2及6-ketoPGFla较Control组显著升高;MgSO_4 7.0、MgSO_4 8.0及MgSO_4 9.0组ET-1、TXA_2较PH组显著下降,而6-keto-PGFla显著升高,其中MgSO_4 9.0组ET-1、TXA_2及6-keto-PGFla较PH组变化最明显。结论 MgSO_4可显著降低肺动脉高压,改善血流动力学紊乱,这与其抑制缩血管因子并促进舒血管因子释放有关。 Objective To investigate the effect of hypertension. Methods Thirty Mongrel dogs were randomly magnesium sulfate for acute pulmonary divided into five groups: control group, pulmonary hypertension (PH) group, MgSO47.0, MgSO48.0, and MgSO49.0 group. The control group was not given any treatment, and the PH group was operated to induce acute pulmonary hypertension; the remaining three groups were induced for acute pulmonary hypertension and after 30 min magnesium sulfate were given through intravenous infusion to maintain the concentration of 7.0 mm/L, 8.0 mm/L and 9.0 ram/L, respectively. The mean pulmonary artery pressure (MPAP) and the circulation mean arterial pressure (MAP) were detected, and endothelin-1 ( ET-1 ), thromboxane A2 ( TXA2 ) and 6-ketoPGFla were measured by immuoflorence method for each group. Results (①The MPAP in PH group were significantly higher than those in control group ; however MPAP in MgSOJ. 0, MgSO48.0 and MgS049.0 were MAP significantly lower, compared with that in PH group, and the lowest level in MgS049.0 group. ②The in PH group was significantly lower than that in control group; however MAP in MgS047. 0, MgSQ8.0 and MgSQ9.0 were significantly higher, compared with that in PH group, and the highest level in MgSO49.0 group. ③ET-1, TXA2 and 6-keto-PGFla in PH group were significantly higher than those in control group. However, ET-1, TXA2 most significantly decreaseds, 6-keto-PGFla increased in MgSO4 9. 0 group compared with those in PH group. Conclusion Magnesium sulfate significantly reduced acute pulmonary hypertension through improvement of circulation disorders by decrement of ET-1 and TXA2 and also increment of prostacyclin( PGI2 ).
