摘要
目的观察苦瓜提取物对自发性高血压大鼠(SHR)血压的影响,并初步探讨其作用机制。方法6~8周龄雄性SHR大鼠50只随机分为模型对照组(SHR组)、卡托普利组(c组)、苦瓜提取物低剂量组(L组)、苦瓜提取物中剂量组(M组)和苦瓜提取物高剂量组(H组),另取10只WKY大鼠作为正常对照组(WKY组)。连续灌胃8周,每2周平行测量大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MBP)。末次给药后腹主动脉取血测定血管紧张素II(AngII)和内皮素(ET-1)等指标。结果与SHR组比较,C组SBP、DBP、MBP极显著降低(P〈0.01),血清AngII和ET-1浓度也显著降低(P〈0.05);L组SBP变化差异无统计学意义,DBP和MBP显著降低(P〈0.05);M组和H组的SBP、DBP、MBP均有不同程度降低(P〈0.01,P〈0.05);H组血清AngII和ET-1浓度显著降低(P〈0.05)。结论苦瓜提取物能降低SHR大鼠的血压,其作用机制可能与降低血清AngII和ET—1浓度有关。
Objective To study the effects of water extracts from Momordica charantia on blood pressure in spon- taneously hypertensive rats ( group SHR) and explore its mechanisms. Methods Fifty 6 - 8 week - old male SHRs were randomly divided into 5 groups: model control group (group SHR), captopril group (group C), water extracts from Momordica charantia in low - dose group ( group L), medium - dose group ( group M) and high - dose group ( group H) . Ten 6 - 8 week - old male wistar - kyoto (WKY) rats were the control group (group WKY ) . Each group were orally adiministered for 8 weeks, and systolic blood pressure ( SBP), diastolic blood pressure (DBP) and mean arterial pressure (MAP) of SHR and WKY was measured every two weeks. At sacrifice, blood was harvested to measure the level of angiotensin II (AngII) and endo- thelin - 1 ( ET - 1 ) in plasma. Results SBP, DBP and MBP in group C was significantly lower than group SHR (P 〈0.01 ), the level of AnglI and ET - 1 was lower than group SHR too (P 〈0.05) . DBP and MBP in group L was lower than group SHR (P 〈 0. 05 ) . SBP, DBP and MBP in group M and group H was lower than group SHR (P 〈0. 01 or P 〈0.05) . The level of AngII and ET- lof group H was lower than group SHR (P 〈 0.05 ) . Conclusions The water extracts from Momordica charantia can depress blood pressure of SHR, and its mechanism may attributed to its lowering effect of plasma AngII and ET - 1 level.
