摘要
目的体外重建异种无细胞真皮活性复合皮。方法将表皮细胞、成纤维细胞种植于异种无细胞真皮基质表面,体外培养约 1周,定时取材行真皮表面 HE染色、常规组织切片 HE染色及扫描电镜观察。结果表皮细胞、成纤维细胞种植后 24 h即贴附于真皮表面,电镜观察可见有伪足形成。培养 5~ 7 d可融合为致密单层细胞膜。结论以无细胞真皮为支架,采用细胞培养技术可于体外重建复合皮。
Objective To reconstitute the living composite graft containing acellular xenodermal matrix(ADM). Methods Keratinocytes or fibroblasts seeded onto ADM were cultured for 1 week,Specimens were harvested at internals,and cells attached to the dermis were observed with the surface of ADM H.E staining and electron microscopy scaning with standard laboratory procedures .Results human keratinocytes and fibroblasts attached well to the dermis within 24h and stretched with pseudopodiums on its surface observed by scanning electron microscopy,and they reached a conflunent monolayer between the 5th and 7d day after seeding.Conclusion A cultured living composite graft composed of keratinocytes or fibroblasts with ADM could be reconstituted in vitro.
