端粒酶与肿瘤治疗研究的新进展

摘要 人的染色体末端包含短的DNA重复序列叫做端粒.端粒的DNA在功能和结构上不同与其他的DNA序列,所有的真核生物,从酵母到人体组织,端粒表现的均是高度保守的重复序列,端粒末端序列的完整性由端粒酶来维持.端粒酶由蛋白质和RNA两部分组成,是一种具有逆转录活性的核糖核蛋白酶,含有引物特异性识别位点,能以自身RNA为模板合成端粒DNA的末端重复序列.由于末端的DNA序列进行性丢失和染色体完整性的逐渐缩短,大多数正常细胞可能被限制分裂.虽然端粒酶靠自身的活性在哺乳动物发育中没有表现出直接的生物学的作用,但在细胞分裂过程中细胞的生存和器官的体内平衡需要靠足够的端粒结构来维持.
