河南商品蛋鸡群马立克氏病病毒流行株的分离与鉴定 被引量:2

Isolation and Identification of Field Strains of Marek's Disease Virus from Henan Province

摘要 2011年~2012年,河南省多个地区养殖场鸡群爆发了马立克氏病,应用外周血淋巴细胞培养分离技术对流行毒株分离并进行了PCR鉴定,最终分离到17个马立克氏病毒毒株,这些病毒株感染鸡胚成纤维细胞3~5d后均能形成典型的病毒蚀斑,从感染细胞总DNA中均能扩增出132bp重复序列的特异性条带,且各毒株间132bp重复序列扩增产物的拷贝数存在明显差异,分离鉴定表明:河南鸡群中可能存在不同毒力马立克氏病毒野毒株的共流行。 Marek's disease( MD) was outbroken in vaccinated chickens in Henan province of China during 2011 ~ 2012. Using chicken embryo fibroblasts( CEF),17 filed isolates were obtained from diseased chickens. The isolates adapted to CEFs very well and most produced typical plaque at 3 ~ 5 d post-infection. Polymerase chain reaction( PCR) amplification of the 132 bp repeats sequence from the viral genome verified that all the isolates are Marek's disease virus( MDV). However,the copies of 132 bp repeats are apparently different in distinct isolates. The filed MDVs of differing virulence may widely spread in the central regions of Henan prorince.
