摘要
目的 观察临床相关浓度利多卡因对过氧化氢损伤红细胞的保护作用。方法 采 2 0例健康人静脉血制成 2 5 %红细胞悬液 ,平均分成空白对照组、过氧化氢 (H2 O2 )损伤组及三个损伤加不同浓度 ( 0 0 1、0 0 2及 0 0 3mg/ml)利多卡因组。孵育后测定红细胞悬液中K+浓度、丙二醛(MDA)浓度和红细胞溶血度。结果 孵育 6 0分钟 ,加药组K+浓度 ( 0 39、0 44、0 35 ppm )、MDA浓度( 7 0 9、6 49、5 98nM )和红细胞溶血度 ( 87 0 4%、83 49%、87 5 1% )与单纯损伤组 (分别为0 5 1ppm、9 19nM和 10 0 0 0 % )相比均显著降低 ,但明显高于空白对照组 ( 0 2 0 ppm、4 13nM、5 2 73% ) ,加药三组间没有显著差异。结论 临床相关浓度的利多卡因对H2 O2 引发的红细胞过氧化损伤有明显保护作用 ,此作用可能是由于减轻了H2 O2 引起的脂质过氧化反应及膜稳定作用而产生。
Objective To determine the protective effect of lidocaine in clinical relevant concentration on the red blood cell(RBC) exposed to hydrogen peroxide(H 2O 2).Methods Intravenous blood samples taken from 20 healthy adults were prepared for RBC suspensions and divided equatly into 5 groups:group Ⅰ as a control;group Ⅱ with H 2O 2(100mM) induced injury;group Ⅲ,Ⅳ and Ⅴ with the same injury as the group Ⅱ after being pretreated with 3 different concentrations of lidocaine(0 01,0 02 or 0 03mg/ml) respectively.The concentrations of potassium and malondialdehyde(MDA) in RBC suspensions and hemolytic degree after incubation were measured respectively.Result After 60 min incubation,the extracellular potassium concentrations(0 39,0 44,0 35ppm),MDA concentrations(7 09,6 49,5 98nM%) and hemolytic degree(87 04%,83 49%,87 51%) were decreased significantly in the groups that pretreated with lidocaine as compared with that in group Ⅱ (0 51ppm,9 19nM and 100 00%) and increased significantly in lidocaine groups than those of the group Ⅰ(0 20ppm,4 13nM,52 73%),but no difference was seen in the lidocaine groups.Conclusion Lidocaine in clinical relevant concentrations may protect RBC from H 2O 2 induced lipid peroxidation injury.
