摘要
目的观察透骨草提取物对人肝癌SMMC-7721细胞增殖的影响,进一步探讨透骨草提取物对SMMC-7721细胞增殖抑制的分子机制。方法MTr法检测透骨草提取物对SMMC-7721细胞增殖的影响,免疫组化方法检测SMMC-7721细胞β-catenin和cyclinD1蛋白的表达。结果6.25—100μg/ml透骨草提取物处理的SMMC-7721细胞增殖抑制率与对照组相比显著增加(P〈0.05),而且随透骨草浓度的增加,抑制率逐渐增加。透骨草提取物对SMMC-7721细胞的半数抑制浓度(IC50)为40.91ug/ml。40.91μg/m1透骨草提取物处理的SMMC-7721细胞β—catenin和cyclinD1的表达明显低于对照组(P〈0.05)。结论透骨草提取物对SMMC-7721细胞的增殖有抑制作用,其机制可能与下调β-catenin和cyclinD1的蛋白表达有关。
Objective To observe the effect of speranskia tuberculata extract on proliferation of human hepatocellular carcinoma SMMC-7721 cells, and to explore its molecular mechanism. Methods The effects of speranskia tuberculata extract on proliferation of human hepatocellular carcinoma SMMC-7721 cells were detected by MTT,and the expression levels of 13-catenin and cyclin D1 protein in SMMC-7721 cells were determined by immunohistochemistry method. Results The inhibition rate on proliferation of SMMC-7721 cells treated by speranskia tuberculata extract at the concentration of 6.25 - 100μg/ml was significantly increased, as compared with that in control group ( P 〈 0.05 ), moreover, with the increase of concentration of speranskia tuberculata extract, the inhibition rate was gradually increased. The ICS0 of speranskia tuberculata extract on SMMC-7721 cells was 40.91 μg/ml. The expression levels of catenin and cyclin D1 in SMMC-7721 cells treated by speranskia tuberculata extract at the concentration of 40.91μg/ml were significantly lower than those in control group ( P 〈 0. 05 ). Conclusion The speranskia tuberculata extract has inhibition effect on proliferation of human hepatocellular carcinoma SMMC-7721 cells, and its action mechanism may be related with down-regulating the expression of β-catenin and cyclin D1 protein.
