摘要
对木薯转基因创新材料进行分子水平的鉴定、评价,通过PCR扩增快速检测,阳性转化材料筛选率为42.9%;Southern Blot检测,阳性材料获得率31.8%,获得了45份木薯转化阳性再生编号材料;半定量RT-PCR检测,目标基因在阳性株系叶片中的表达与野生型差异不大,而在根中的表达与野生型相比有明显差异;并观测、记录、评价其表型变异特征。说明建立了可靠的遗传转化再生体系,为今后木薯育种奠定良好的技术与材料基础。
By the molecular techniques of identification and evaluation transgenic innovative materials in cassava (Manihot esculenta crantz) ,rapid detection by PCR, the positive rate of transformation materials was 42.9%, detection by Southern Blot, obtained rate of positive material was 31.8%, 45 regeneration materials were got.Detection by Sem-quantitative RT-PCR,the target gene expression approximate in leaves with the wild type,while the expression in roots had significant difference.Observation, recording, evaluation of phenotypic variation positive materials were carried out.In short, a reliable system for genetic transformation regeneration was established, which provided technical and material foundation for cassava breeding.
