摘要
应用双链RNA(dsRNA)诱导的RNA干扰(RNAi)技术原理,将体外合成的源于白背飞虱肌动蛋白(Actin)基因的dsRNA通过膜饲喂法饲喂其2龄若虫.通过优化人工饲料、dsRNA浓度和饲喂时间,成功地干扰了白背飞虱Actin基因的表达,并引发白背飞虱的死亡,从而建立了白背飞虱体内的RNAi体系.
The potential of RNA interference (RNAi) induced by double-strand RNA (dsRNA) was tested in white-backed plan- thopper (WBPH). dsRNAs corresponding to the Actin gene of WBPH were produced in vitro and applied to the 2nd instar nymphs of WBPH through a membrane-feeding method. After optimizing the artificial diet, dsRNA concentration and feeding time, the ex- pression of Actin in cells of WBPH was significantly reduced, leading to death of the insects prematurely. This indicated that the in vivo RNAI system for WBPHs was established.
