摘要
目的探讨选择性多巴胺D4受体激动剂ABT-724对注意缺陷多动障碍(ADHD)动物模型!自发性高血压大鼠(SHR)注意定势转移能力的影响。方法根据ABT-724的用量(腹腔注射,0.04、0.16、0.64 mg/kg),将SHR大鼠分为低、中、高剂量组,以哌甲酯组、SHR盐水组及WKY盐水组为对照,采用行为观测的方法,观察该药物对SHR大鼠注意定势转移能力的影响。结果在注意定势转移实验中,低、中、高剂量组与SHR盐水组相比,其循环数、错误数均降低(P<0.01);与哌甲酯组相比,差异无统计学意义(P>0.05)。错误类型分析结果显示,低、中、高剂量组与SHR盐水组比较,固执错误数和退化错误数均明显降低(P<0.01),而非强化错误数无改变(P>0.05);与哌甲酯组相比,差异均无统计学意义(P>0.05)。结论 ABT-724可有效改善SHR大鼠的注意定势转移能力,而且与哌甲酯的效果相当。
Objective To investigate the effects of ABT-724, a selective dopamine D4 receptor, on attentional setshifting behaviors in spontaneously hypertensive rats ( SHR), a rat model of attention-deficit hyperactivity disorder (ADHD). Methods According to the dose of ABT-724 ( intraperitoneal injection, 0.04, 0. 16 and 0.64 mg/kg), SHR rats were divided into three ( low, medium and high dose) groups. SHR rats intervened with methylphenidate (25 mg/kg) or saline and Wistar-Kyoto (WKY) rats intervened with saline were all taken as control groups, and they were named as methylphenidate group, SHR saline group and WKY saline group, respectively. The effects of ABT-724 on attentional set-shifting behaviors in SHR rats were observed. Results Compared with SHR saline group, the trials and errors to reach criterion were all reduced in low, medium and high dose group (P 〈 0.01 ). Compared with meth- ylphenidate group, there was no obvious difference in trials and errors to reach criterion for all the three dose groups ( P 〉 0.05 ). According to the error types, low, medium and high dose groups showed less perseverative and regressive errors than SHR saline group ( P 〈 0.01 ), but showed no obvious difference in never-reinforced error ( P 〉 0.05 ). Compared with methylphenidate group, there was no significant change in error types for all the three doses groups ( P 〉 0.05 ). Conclusion ABT-724 can improve attentional set-shifting behaviors in SHR rats as methylphenidate.
