摘要
采用二次通用旋转组合设计实验优化超声波辅助碱性蛋白酶提取杜仲籽粕蛋白工艺。在单因素实验基础上,选择pH值、碱性蛋白酶用量、酶解温度与酶解时间为考察因素,以杜仲籽粕蛋白提取率为响应值建立回归数学模型,对杜仲籽粕蛋白提取工艺条件进行优化。结果表明,最佳提取工艺条件为:pH 10,碱性蛋白酶用量320 U/g,酶解温度55℃,酶解时间115 min。在此优化条件下,杜仲籽粕蛋白提取率可达74.28%。
The technological condition tbr enzymatic extraction of Eucommia seed meal protein was optimized by using alka- line pro|ease wilh quadratic regression rotational composite design and ultrasonic-assisted. Based on the single factors, pH, dosage of alkaline prolease, enzymatic extraetion temperature amt enzymatic extraction time, were selected as experiment factors. The regression mathematics model was established with extraction efficiency as criteria. The optimization extraction conditions were achieved throt, gh the results of variance analysis and mathematics model optimization. The optimum extrac- tion conditions were obtained as follows: extraetion temperature of 55%, time of 115 min, pH 10 and dosage of alkaline pro- tease of 320U/g. Under this condition, extraction efficiency of Eucommia seed meal protein can be reached to 74.28%.
