摘要
目的探讨TMEM16A氯通道在食管鳞状细胞癌发生发展中的意义。方法运用RT-PCR、Westernblot方法分别从mRNA水平、蛋白水平检测食管鳞状癌细胞TE-1细胞株中TMEM16A的表达;运用免疫组织化学PV法,检测食管癌组织TMEM16A蛋白的表达。结果 TE-1细胞株及食管癌组织均表达有TMEM16A通道,并且免疫组化结果表明不同分化程度的食管鳞癌组织中,高、中、低分化程度的阳性表达率分别为38.5%、62.5%、90%,三者比较差异有显著性(P<0.05)。TMEM16A在淋巴结转移组中的阳性表达率明显高于无淋巴结转移组(71.4%vs 22.2%,P<0.05)。结论 TMEM16A的表达可能在肿瘤发生发展过程中发挥了一定的作用,TMEM16A在食管癌组织中的表达有望为食管癌的分子诊断和基因治疗提供新的靶点。
[Objective] To study the expression of TMEM16A in esophageal squamous cell carcinoma (ESCC), and to explore its potential role in carcinogenesis and progression of esophageal cancer. [ Methods ] RT-PCR and western-blot were used separately to detect the expression of TMEM16A in TE-1 cell line. PV method was used to identify the expression of TMEM16A in ESCC. [Results] TE-1 cell line and ESCC both expressed TMEM16A chloride channel. And immuno-histochemistry resuhs showed that the positive rate of TMEM16A in high, middle and low differentiation groups were 38.5%, 62.5% and 90% respectively and there was significant statistically difference (P 〈0.05). The positive rate of TMEM16A in lymph node positive group was higher than that in lymph node negative group and there was significant statistically difference (71.4% vs 22.2%, P 〈0.05). [ Conclusions ] TMEM16A may play an important role in carcinogenesis of ESCC. TMEM16A is expected to be a new biomarker in molecular diagnosis and gene therapy for ESCC.
