RhoA和Ezrin蛋白在食管鳞癌中的表达及临床意义 被引量:1

摘要 目的检测RhoA和Ezrin蛋白各自在不同临床分期食管鳞癌组织中的表达,探讨两者在食管鳞癌发生、侵袭和转移中的作用以及它们在其中的相互关系。方法应用免疫组织化学SABC法检测95例食管鳞癌组织及其相应癌旁正常食管黏膜上皮组织中RhoA和Ezrin蛋白的表达水平,并结合临床相关病理特征进行统计学分析。结果 RhoA和Ezrin蛋白在食管鳞癌组织中的表达率分别为69.5%和72.6%,在癌旁正常食管黏膜上皮组织中表达率分别为11.7%和12.6%,两组病例间的两种蛋白表达两两比较均有显著性差异(P<0.05)。RhoA和Ezrin蛋白表达水平与食管鳞癌患者的年龄、性别、饮酒与否、肿瘤部位、分化程度无关(P>0.05),而与肿瘤浸润程度、淋巴结转移、TNM分期密切相关(P<0.05)。食管鳞癌组织中RhoA和Ezrin蛋白表达呈正相关(Kappa系数=0.770,P<0.05)。结论 RhoA和Ezrin蛋白均在食管鳞癌的侵袭和转移过程中起重要作用,提示二者在该过程中可能起着协同作用。 [ Objective ] To investigate the expression of RhoA and Ezrin proteins in different clinical stages of esophageal squamous cell carcinoma, and analyze their roles and mutual relations in tumorigenesis, invasion and metastasis. [Methods] The expressions of RhoA and Ezrin in 95 specimens of primary esophageal squamous cell carcinoma, 95 specimens of paired adjacent normal esophageal tissues were detected by immunohistochemical SABC staining. The results were correlated with elinieopathological parameters. [ Results ] The positive expression rates of RhoA in normal esophageal tissues and esophageal squamous cell carcinoma tissues were 11.7% and 69.5%, and those of Ezrin were 12.6% and 72.6%, respectively. The expressions of RhoA and Ezrin were significantly correlated to depth of invasion(T), lymph node metastasis(N), clinical TNM stage, respectively (P 〈0.05), but not to age, gender, drinking, tumor location, tumor grade (P 〉0.05). The expression of RhoA was positively correlated to that of Ezrin in esophageal squamous cell carcinoma (P 〈0.05). [ Conclusions ] Our findings suggested that high expressions of RhoA and Ezrin might have played important roles in the malignant progression of invasion and metastasis of esophageal squamous cell carcinoma, which indicated that they might have played synergistic effects during the process of invasion and metastasis.
