摘要 目的通过检测mTOR在子宫肌瘤、肌瘤旁正常平滑肌组织和正常子宫平滑肌组织中mRNA和蛋白的表达,探讨mTOR与子宫肌瘤发病的关系。方法选子宫肌瘤患者25例取肌瘤中心部位及远离瘤体正常平滑肌组织为肌瘤组和肌瘤旁组,选因宫颈上皮内瘤样病变Ⅲ级行子宫切除术患者25例取正常子宫平滑肌组织为非肌瘤组。通过RT-PCR、免疫组织化学法检测三组标本mTOR在mRNA及蛋白水平的表达差异。结果肌瘤组mTOR mRNA及蛋白水平表达强度较肌瘤旁组和非肌瘤组均明显升高(P<0.01);肌瘤旁组与非肌瘤组的表达强度差异无显著性(P>0.05)。mTOR在肌瘤组、肌瘤旁组和非肌瘤组中mRNA和蛋白表达强度在增生期和分泌期无差异(P>0.05)。结论 mTOR在子宫肌瘤组织中表达明显增强,提示mTOR可能参与子宫肌瘤的发病过程。 [ Objective ] To investigate the expression of mTOR (mammalian target of rapamycin, mTOR) and its role in the pathogenesis and development of uterine myoma. [ Methods ] 25 patients with uterine myoma who underwent hysterectomy were selected for uterine myoma group and the adjacent hysteromyoma group. Another 25 patients underwent hysterectomy because of cervical intraepithelial neoplasia m were also selected for non-hysteromyoma group. Immunohistochemieal methods and reverse transcription polymerase chain reaction were used to examine the expression of mTOR and the expression level was tested by image analysis system. [ Results ] The expression level of mTOR mRNA and protein inn uterine myoma group was obviously higher than that of adjacent hysteromyoma group or that of none-hysteromyoma group (P 〈0.01). But no significant difference was observed in the expression level of mTOR mRNA between adjacent hysteromyoma group and non-hysteromyoma group (P 〉0.05). There was no significant difference in the expression level of mTOR mRNA and protein in each group between proliferative phase and secretory phase. [Conclusions] The expression level of mTOR was higher in uterine myoma group than that in adjacent hysteromyoma group and non-hysteromyoma group, which suggested mTOR may involve in the pathogenesis of uterine myoma.
