摘要
目的探讨野生型(wt)-p53与JunB融合基因抑制裸鼠肝细胞肝癌(肝癌)移植瘤的作用。方法将18只BALB/c裸鼠按随机数字表法随机分为实验组、阴性对照组和空白对照组,每组6只,分别接种wt-p53与JunB融合基因质粒、携带GFP的空质粒及单纯肝癌细胞HepG2,于注射后的28d处死荷瘤裸鼠,测量肿瘤体积、肿瘤重量,计算肿瘤抑制率。结果 3组裸鼠移植瘤位于背侧皮下,实验组裸鼠移植瘤较小,阴性对照组与空白对照组裸鼠移植瘤均明显隆起于裸鼠背部皮下。实验组移植瘤的体积、重量明显小于阴性对照组,差异均有统计学意义(LSD-t=4.63,P=0.006;LSD-t=19.56,P<0.01);实验组移植瘤的体积、重量也明显小于空白对照组,差异均有统计学意义(LSD-t=5.74,P=0.002;LSD-t=27.22,P<0.01);阴性对照组与空白对照组间移植瘤的体积、重量差异无统计学意义(LSD-t=1.59,P=0.171;LSD-t=2.00,P=0.101)。实验组平均肿瘤抑制率为84%。结论 wt-p53和JunB融合基因可明显抑制裸鼠肝癌移植瘤的生长。
Objective To examine the inhibitory effect of fusion gene of wild-type p53 and JunB on the growth of transplanted hepatocellular carcinoma in nude mice.Methods Eighteen nude mice were randomly divided into experiment group,negative control group and blank control group(n=6each).They were subcutaneously injected with hepatocellular carcinoma cells transfected with plasmids carrying wt-p53/JunB fusion gene or empty plasmids carrying GFP,or hepatocellular carcinoma cells alone.Twenty-eight days after the injection,the nude mice were sacrificed and the gross tumor volume,the tumor weight and the tumor inhibition rate were obtained.Results The hepatocellular carcinomas were subcutaneously transplanted on the back of the mice.They were significantly protruded in the negative and blank control groups compared with the experiment group.The gross tumor volume and tumor weight were significantly decreased in the experiment group relative to the negative control group(LSD-t=4.63,P=0.006;LSD-t=19.56,P0.01)and the blank control group(LSD-t=5.74,P=0.002;LSD-t = 27.22,P0.01).But there was no significant difference in the gross tumor volume and tumor weight between the negative control group and the blank control group(LSD-t=1.59,P=0.171;LSD-t= 2.00,P=0.101).The tumor inhibition rate of the experimental group was 84%.Conclusion The fusion gene of wt-p53and JunB can significantly inhibit the growth of the transplanted hepatocellular carcinoma in nude mice.
