急性血糖波动对心肌细胞内炎症因子的影响

摘要 目的探讨急性血糖波动对心肌细胞内白细胞介素(IL)-6、肿瘤坏死因子(TNF)-α水平及细胞间黏附分子-1(ICAM-1)表达的影响。方法以间断静脉输注50%葡萄糖注射液48h的方法,建立急性血糖波动和高血糖动物模型。采用酶联免疫吸附试验(ELISA)法检测IL-6及TNF-α水平。免疫组织化学的方法检测心肌细胞中ICAM-1表达。结果急性血糖波动组心肌细胞中IL-6、TNF-α水平及ICAM-1表达显著升高。结论急性血糖波动能导致机体的炎症因子水平明显升高,从而损伤心肌。 Objective To investigate whether acute blood glucose fluctuation has the effect on the level of interleukin-6 (IL-6), tumor necrosis factor-~ (TNF-a) and the expression of intercellular adhesion molecule-1 (ICAM-1). Methods The Wistar rats were infused intermittently or consistently with 50% glucose solution. The level of IL-6 and TNF-a was investigated through ELISA. The expression of ICAM-1 was investigated through the method of immunohistochemisty. Results The levels of IL-6 and TNF^a in the acute blood glucose fluctuation group were evidently higher than that of consistently high blood glucose group and the control group (P^0.05). The expression of ICAM-1 in the cytoplasm in myocardial cells of acute blood glucose fluctuation group was evi- dently increased than that in other two groups (P^0.05). Conclusion The level of inflammatory factors induced by acute blood glucose fluctuation was apparently higher, which causes the myocardium injury.
