百事乐胶囊对慢性应激抑郁大鼠海马BDNF和TrkB表达的影响

Effects of Baishile Capsules on Expression of Brain-derived Neurotrophic Factorand Tropomyosin Receptor Kinase Bin Hippocampus of Chronic Stress Depression Rats

摘要 目的观察百事乐胶囊对慢性应激抑郁大鼠海马脑源性神经因子(BDNF)和酪氨酸激酶B(TrkB)表达的影响。方法 SD大鼠随机分为正常组、模型组、百事乐组和氟西汀组,每组8只。除正常组外,其他组先给予21 d慢性不可预见性应激造模,然后在应激前1 h灌胃给药21 d,其中模型组大鼠灌胃蒸馏水,氟西汀组灌胃盐酸氟西汀药液,百事乐组灌胃百事乐胶囊药液。旷场实验检测大鼠行为学变化,免疫组化检测大鼠海马BDNF和TrkB的表达。结果与模型组比较,百事乐胶囊能显著改善大鼠的水平活动和垂直活动水平(P<0.05或0.01),提高海马BDNF和TrkB的表达(P<0.01)。结论百事乐胶囊可调节海马BDNF和TrkB表达,是其促进抑郁大鼠海马神经功能恢复的机制之一。 Objective: To investigate the effects of Baishile Capsules (BSL) on the behaviors and brain-derived neurotrophic factor (BDNF) and tyrosine kinase receptor B (TrkB) in hippocampus of chronic stress depression rats. Methods: SD rats were randomly divided into the healthy control group (HCG),model group(MOG),BSL group(BSG) and fluoxetine group(FYG). Besides the I-ICG,all the animals in each group were exposed to chronic stress with 21 days for model ,and then they were respectively given the corresponding medicine before modeling. MOG were given the same value of distilled water. Body weights of rats were measured. The behavioral indices were tested by open-field experiment. The expressions of BDNF and TrkB in hippocampus were measured by im- munohistochemical staining. Results: Compared with MOG,the horizontal and vertical activitiesof BSG were im- proved obviously(P〈 0.05 or 0.01 ) and the expression of BDNF and TrkB of BSG were enhanced remarkably(P〈 0.01). Conclution: Baishile Capsules could regulate the expression levels of BDNF and TrkB which may be one of the anti-depression mechanism with the recovery of neurological function.
