摘要
目的研究人乳腺癌细胞中TRIM22对NF-κB表达的影响。方法通过RT-PCR和Western blot方法检测人乳腺癌细胞系MCF-7和人正常乳腺细胞系MCF-10A中TRIM22和NF-κB在mRNA和蛋白水平的表达差异;构建TRIM22表达质粒,并通过瞬时转染TRIM22表达质粒,使MCF-7高表达TRIM22,再检测NF-κB在mRNA及蛋白水平的变化。结果TRIM22在人正常乳腺细胞MCF-10A中的表达水平高于乳腺癌细胞系MCF-7,在MCF-7中转染TRIM22表达质粒后,NF-κB的表达量随之降低。结论在MCF-7细胞中TRIM22可负调控NF-κB的表达。
Objective To investigate the effect of TRIM22 on the expression of NF-KB. Methods RT-PCR and Western blot were performed to detect the expression of TRIM22 and NF-xB in MCF-10A cells and MCF-7 cells, respectively. TRIM22 expression plasmid was constructed with Xho I and Hind 111. Via the transient transfection, the level of over-expression of the TRIM22 from MCF-7 cells was increased and then the mRNA and protein expression of TRIM22 were detected,respectively. Results Both RT-PCR and West- ern blot indicated that the level of TRIM22 expression in normal human mammary cell line MCF-10A was higher than that in breast cancer cell line MCF-7. Expression of NF-KB decreased in MCF-7 transfected with the TRIM22 expression plasmid. Conclusions TRIM22 can down-regulate the expression of NF-KB in MCF- 7 ceils.
