尖锐湿疣患者肿瘤易感基因的基因芯片技术检测 被引量:2

摘要 目的分析尖锐湿疣肿瘤易感基因,为筛查和判断尖锐湿疣预后提供理论依据。方法选择5例尖锐湿疣,3例鳞癌和3例正常包皮标本,提取DNA,采用全基因组外显子测序技术,分析疣组织的肿瘤易感基因。结果 6号染色体上发现10个与肿瘤发生相关的单体型(P<0.001)、18个相关基因。查询GSE9750数据集,该数据集显示上调的有TRIM39,HCP5,C2(P<0.05);下调的有DOM3Z,STK19(P<0.05);差异不明显的有MOG,C6orf47,APOM,CFB,SKIV2L,FKBPL和ATF6B(P>0.05)。结论TRIM39,HCP5,APOM,C2,DOM3Z,STK19,FKBPL可能为尖锐湿疣患者癌变的易感基因。 Objective To analyze tumor susceptibility genes in patients with condyloma acuminatum and provide theo- retical basis to screening and judging prognosis of condyloma acuminatum. Methods Five cases of condylo- ma acuminatum, 3 cases of squamous carcinoma and 3 cases of normal prepuce specimens were included in this study. DNA was extracted and the whole genome exon sequencing was carried out to analyze tumor sus- ceptibihty genes of wart tissue. Results Ten haplotyp blocks related to tumorigenesis and 18 genes related tumorigenesis on chromosome 6 was found( P 〈0.001 ). We compared our results with the dataset GSE9750. The up-regulated genes included TRIM39, HCP5, C2 ( P 〈 0.05 ) ; The down-regulated included DOM3Z, STK19(P 〈0.05). Conclusion TRIM39, HCP5, APOM, C2, DOM3Z, STK19, FKBPL may be the cer- vical toumor susceptibility genes in the patients with condyloma acuminatum.
