九节龙皂苷诱导胶质瘤细胞SHG-44MG发生自噬性细胞死亡及其机制研究 被引量:4

摘要 目的研究九节龙皂苷(A·DS)诱导人脑胶质瘤细胞SHG-44MG细胞发生自噬性细胞死亡及其机制研究。方法 MTT法检测A·DS对人胶质瘤细胞SHG-44MG增殖的影响,Western blot定量分析自噬相关蛋白Beclin-1、LC-3的表达。免疫细胞化学定位、定性检测自噬相关蛋白Beclin-1、LC-3的表达。结果随着A·DS浓度的增加,SHG-44MG细胞的生存率降低;Western blot结果表明,随着A·DS浓度的增大,SHG-44MG细胞中的自噬相关蛋白Beclin-1、LC-3的表达明显增加。免疫组化实验结果显示,A·DS可剂量依赖性的促进自噬相关蛋白Beclin-1、LC-3的表达。结论 A·DS可剂量依赖性的抑制SHG-44MG细胞增殖,通过促进自噬相关蛋白Beclin-1、LC-3的表达,诱发SHG-44MG细胞发生自噬性细胞死亡。 Objective To study the effect of ardipusilloside on human glioma SHG-44MG cells and its mechanism. Methods MTT assay was performed to measure the effect on the proliferation of SHG-44MG cells treated with different concentrations of ardipusilloside for 24,48,and 72 h. The autophagy of SHG-44MG cells was detected by Western blot. Beclin-1 and LC-3 were tested by immunocytochemistry. Results Ardipusilloside significantly inhibited the viability of SHG-44MG cells in a concentration and time-dependent manner. Western blot showed that Beclin-1 and LC-3 were significantly increased with the dose. Immunocytochemical results showed that Beclin-1 and LC-3 were highly expressed after treatment with ardipusilloside. Conclusion Ardipusilloside can effectively induce the autophagy of glioma SHG-44MG cells was due to the activation of Beclin-1 and LC-3.
