基因表达谱芯片筛选寻常型银屑病相关基因的研究 被引量:1

摘要 目的 运用基因芯片技术研究寻常型银屑病的基因表达谱,筛选寻常型银屑病相关基因.方法 从4例寻常型银屑病患者及1例正常人的外周血中提取并分离得到总RNA.将总RNA纯化,并逆转录合成单链、双链cDNA.纯化双链cDNA后,体外转录合成生物素标记的cRNA,并与寡核苷酸芯片进行杂交.使用激光共聚焦扫描仪进行图像扫描,配合数据处理软件读取、处理数据,筛选出2倍及以上差异表达基因.结果 筛选出的2倍及以上差异表达基因占每张芯片上基因总数的1.270%~4.644%,这些基因主要分属于免疫相关基因、细胞信号和传递蛋白相关基因、代谢相关基因、蛋白质翻译和合成相关基因、DNA结合、转录和转录因子类基因、发育相关基因等.4例寻常型银屑病患者与正常人相比较有共同差异表达的基因共52个,其中17个基因表达上调,35个基因表达下调.结论 基因芯片技术有较高的重复性和稳定性,能有效地进行寻常型银屑病相关基因的初步筛选. Objective To investigate the gene expression profiling of psoriasis vulgaris with Gene chip technology, and to screen the associated genes of psoriasis vulgaris. Methods The total RNA was extracted from the periphery blood samples of 4 psoriasis vulgaris pa- tients and 1 normal control. The total RNA was purified and reversely transcripted to single-stranded and double-stranded cDNA. The biotin labeled cRNA was produced by transcription and hybridizated with oligonucleotide microarray in vitro. Image scanning was per- formed using a confocal laser scanner. Screening out 2 times and above differentially expressed genes with Data processing software. Re- suits The numbers of 2 times and above differentially expressed gene were accounted for the total number of genes 1. 270% - 4. 644% in each chip. These genes mainly related to the immunity, the cell signal, the transmission protein, the metabolism, the protein transla- tion and synthesis, the DNA combination and transcription and transcription factor, the growth, and so on. Compared with the control, 52 genes that common differentially expressed were picked up from 4 samples of psoriasis vulgaris. Among 52 genes, 17 genes were up- regulated and 35 genes were down-regulated. Conclusion The gene chip technology has the high duplication and the stability. It can ef- fectively carry on the preliminary screening of psoriasis vulgaris associated genes.
