黄瓜籽乙酸乙酯提取物对人成骨肉瘤细胞骨形态发生蛋白-2的影响 被引量:5

摘要 目的通过测定骨形态发生蛋白-2(BMP-2)的表达,探讨黄瓜籽乙酸乙酯提取物(CSA)对人成骨肉瘤(MG63)细胞的活性的影响。方法采用体外培养MG63细胞,给予CSA与MG6置于37 e,5%CO2饱和湿度培养箱中共同孵育48 h,MTT比色法检测黄瓜籽提取物对细胞活性的影响;生化法检测ALP含量,ELISA法定量检测BMP-2的含量;免疫组化法测定BMP-2表达。结果 MTT法,CSA具有明显促进MG63细胞活力(P<0101);免疫组织化学染色法,CSA细胞中的BMP-2的表达量明显高于空白组;CSA组ALP和BMP-2含量高于空白组(P<0101)。结论 CSA对MG63具有保护作用,能够有效的提高MG63细胞的生存率,其机制可能与促进BMP-2表达、升高ALP和BMP-2的含量有关。 Objective To evaluate the expression of bone morphogenetic protein (BMP-2) of ethyl acetate extract of cucumis sativus seed on the Human Osteosarcoma MG63 ceils. Method MC,63 cells with CSA were in vitro at 37 ℃ and 5 % CO2 saturated humidity incubator in common incubation 48 h. Cell viability was measured by MTT assay, and expression of BMP-2 was performed by the method of immunohistochemistry and ELISA. ALP was detected by Biochemical method. Result CSA can obviously promote MG63 cells vitality( P 〈 0.01 ) by MTT. The amount of the expression of BMP-2 in the CSA group wasobviously higher than that of blank group by Immunohistochemical staining. ALP and BMP-2 level of CSA group were higher than that of blank group ( P 〈 0.01). Conclusion CSA has a protective effect and can effectively improve the cell survival. Its mechanism may be related to promotion of the expression of BMP-2 and increase of the content of ALP and BMP-2.
