全自动血细胞分析仪与手工计数法测定嗜酸性粒细胞的比较

摘要 目的评价MEK-8222J/K型和BeckmanCoulterLH750型全自动血细胞分析仪对嗜酸性粒细胞测定的临床应用价值。方法选取湖北省中医院2012年2月~7月住院患者100例,用直接计数法、间接计数法、MEK-8222J/K型和BeckmanCoulterLH750型血细胞分析仪分别对其中3份标本进行精密度测定,并对所有标本进行准确度测定。结果直接计数法、间接计数法、MEK-8222J/K型和BeckmanCoulterLH750型细胞分析仪对3份标本的测定结果的批内平均CV%分别为6.66%,7.78%,5.83%和4.79%。间接计数法、MEK-8zzzj/K型和BeckmanCoulterLH750型血细胞分析仪对100份标本的测定结果与直接计数法经配对t检验,t值分别为1.125,0.463和0.116,均小于t0.05.100,P〉0.05,三种方法与直接计数法的差异均无统计学意义。结论实验结果表明MEK-8222J/K型和BeckmanCoulterLH750型血细胞分析仪对嗜酸性粒细胞的分析具有良好的准确度和精密度,并且检测速度快。因此,嗜酸性粒细胞在检出限以上时可以用全自动血细胞分析仪替代手工计数法进行测定。 Objective To study the clinical value of eosinophil granulocyte count detected by MEK-8222J/K and Beckman Coulter LH750 automatic blood cell analyzer. Methods 100 cases of blood samples collected from hospitalized patients in Huhei provincial hospital of TCM during February to July in 2012. The precision determination of 3 samples eosinophil gran- ulocytes each detected by MEK-8222J/K, Beckman and Coulter LH750 automatic blood cell analyzer, directly count and indi- rectly count,all samples' accury detected. Results The 3 samples' average CV% detected by MEK-8222J/K, Beckman and Coulter LH750 automatic blood cell analyzer in direct count, indirect count method were 6. 66%, 7.78%, 5.83% and 4. 79 %, respectively. Copmared the results of 100 samples detected by indirectly count, MEK-8222J/K, Beckman and Coulter LH750 automatic blood cell analyzer with directed counting method by t-test,t were 1. 125,0. 463 and 0. 116,all less than to. 0s,100, P〉0. 05. There was no significant difference between the three methods and direr counting method. Conclusion The accuracy and precision of MEK-8222J/K,Beckman and Coulter LH750 automatic blood cell analyzer in eosinophil granulo- cyte counting were favorable, and counting speed were fast as well. Thus, Automatic blood analyzer is recommended to detect eosinophil in place of counted by hand when eosinophil granulocytes are beyond the limit detection.
