摘要 目的 :采用计算机优化技术优化甲氨蝶呤人血白蛋白微球的制备工艺。方法 :以均匀设计方法考察不同因素在3个水平下对微球平均粒径的影响。实验结果以多元线性回归法处理。结果 :白蛋白浓度和乳化搅拌速度对微球大小影响显著。乳化时间和乳化剂用量对微球大小没有显著影响。结论 :计算机优化技术可以有效优化微球制备工艺 。 OBJECTIVE:A computer optimization technique was applied to the design of preparation technique of methotreate human serum albumin microspheres.METHODS:The effects of different factors on mean diameter of microsphere was studied at three levels accoring to uniform design of experiment.Multivariable linear regression was used to determine the relationship between the factors applied with the effects responsed.RESULTS:Albumin concentration and stirring rate during emulsification had significant effect on mean diameter of microspheres and emulsification time and the amount of albumin had no significant effect.CONCLUSION:Computer optimization technique can optimize preparation technique of microspheres effectively.The method has potentiality to be use in other dosage forms.
