温度对LPS诱导人单核细胞THP-1活性的影响

摘要 目的:研究不同温度对LPS诱导人单核细胞系(THP-1细胞)体外活性的影响。方法:在不同温度培养条件下用LPS刺激人THP-1细胞,分别检测刺激上清中TNF-!、IL-1β及NO的表达量,并采用定量RT-PCR方法对TNF-!和IL-1βmRNA表达量进行检测。结果:在40℃培养条件下,LPS刺激THP-1细胞产生TNF-!、IL-1β及NO的水平要早于37℃及34℃培养条件下。此外,在所有LPS刺激组,TNF-!及IL-1βmRNA在刺激1小时后就大量表达,尤其在40℃条件下。但同样在此条件下,LPS刺激12小时后TNF-!及IL-1βmRNA表达量急剧降低,而37℃及34℃培养条件下依然保持较高水平。结论:本研究说明了培养温度对LPS刺激THP-1细胞产生前炎症介质的活性有显著影响。 Objective:To investigate the effect of different temperature on the kinetics of lipopolysaccharide (LPS)-induced human THP-1 cell activity in vitro. Methods: Human THP-1 cells were stimulated with LPS at different temperatures (34℃, 37℃ and 40℃ ), then, TNF-α, IL-1β and NO productions of stimulant supernatant were measured. Also, Levels of TNF-αand IL-1β mR- NA were measured by RT-PCR. Results: Early elevation of TNF-α IL-1 βand NO productions were found in LPS-stimulated THP-1 ceils, the highest level was incubated at 40℃ ,followed by ceils that incubated at 37℃ and the lowest level was detected at 34℃. Ex- pression of TNF-αmRNA and IL-1 β mRNA were observed as early as 1 h post exposure to LPS in all studied temperatures, but de- creased sharply after 12 h post exposure to LPS in LPS-stimulated THP-1 cell that incubated at 40℃ only. Conclusion: The study de- scribes the striking effects of incubation temperature on activity of LPS-stimulated THP-1 cell.
