摘要
选择ITS、ITS2、psbA-trnH、matK 4条序列进行评价,以扩增及测序成功率、变异位点数和k-2-p距离等指标评价各序列的鉴定能力。此外,基于MEGA5分析不同品种美国山核桃序列种间K2P遗传距离并构建NJ树。结果显示:不同品种美国山核桃4条序列的扩增及测序成功率扩增均为100%。不同品种的ITS2和psbA-trnH序列一致性为100%,而ITS和matK序列的一致性分别为99.32%和99.95%,两者均存在较多的变异位点,可以有效地区分美国山核桃不同品种。因此,ITS和matK序列可以作为鉴定美国山核桃不同品种的优势条形码组合。
The ITS2, psbA-trnH, ITS and matK sequences were used for identification of pecan varieties. The PCR amplification and sequencing efficiency, variation sites and k-2-p distance were employed to evaluate their capa- bilities of identification. In addition, K2P distances were calculated and NJ Tree was constructed. The result showed that the amplification and sequencing efficiencies of all pecan varieties were 100.0%. The identity of ITS2 sequence in pecan varieties were 100.0% , and so did psbA-trnH sequence. The similarities of ITS and matK in pecan varieties were 99.32% and 99.95% , respectively. Many variation sites in ITS and matK could help to identify pecan varieties efficiently.
