雷公藤甲素在调控人鼻咽癌转移相关基因中的作用及机制 被引量:3

摘要 目的探讨雷公藤甲素在调控人鼻咽癌转移相关基因中的作用及机制。方法人鼻咽癌CNE1细胞经雷公藤甲素处理后,通过Western blot试验考察肿瘤细胞E-cadherin、MMP-2/9及p-Akt蛋白表达的变化,Real-time PCR试验考察肿瘤细胞E-cadherin和MMP-2/9 mRNA表达的变化,报告基因试验考察NF-κb启动子活性的变化。结果 CNE1细胞经过5和10μmol·L-1雷公藤甲素处理24 h后,MMP-2/9及p-Akt蛋白表达显著下降,E-cadherin蛋白表达显著上调;mRNA表达MMP-2分别下降69.1%和37.4%(P<0.05),MMP-9分别下降83.2%和35.2%(P<0.05),E-cadherin分别上调132.8%和155.6%(P<0.05),NF-κb启动子活性分别下降51.2%和34.8%(P<0.05)。结论雷公藤甲素可抑制人鼻咽癌CNE1人鼻咽癌转移相关基因表达,其机制可能与其下调细胞NF-κb启动子活性有关。
