摘要
目的 建立同时测定半夏中尿苷、鸟苷和腺苷含量的反相高效液相色谱(RP-HPLC)梯度洗脱法.方法 采用ZORBAX-SB-C18色谱柱(150 mm ×4.6 mm,5 μm);以水-甲醇为流动相,梯度洗脱,流速为0.6 mL·min-1,柱温为25 ℃,检测波长260 nm.结果 尿苷在0.078 4~0.784 0 μg范围内线性关系良好,r=0.999 5;鸟苷在0.079 8~0.798 0 μg范围内线性关系良好,r=0.999 5;腺苷在0.085 2~0.852 0 μg范围内线性关系良好,r=0.999 4.结论 该方法操作简便,方法可靠,重复性好,可以作为半夏的质量控制方法之一.
Objective To establish a RP-HPLC gradient elution method for detetxnination of uridine, guanosine and adenosine in Pinellia ternate. Methods The extracts were separated on a ZORBAX-SB-CIs analytical column (4.60 mm×150 mm,5 μm) with a solvent system containing CH3OH-H20, the flow rate at 0.6 mL· min-1 and the column temperature at 25 ℃. The analytes were detected at 260 nm UV wavelength. Results The linear ranges of uridine, guanosine and adenosine were 0.078 4-0.784 0 μg(r=0. 999 5), 0.079 8-0.798 0 μg(r= 0.999 5), and 0.085 2-0.852 0 μg(r=0.999 4), respectively. Conclusion This method has a good reproducibility, which can be used for quality control of Pinellia ternate.
