丹参不同组分对非酒精性脂肪肝大鼠肝脏PPARαmRNA表达的影响 被引量:9

摘要 目的:观察中药丹参的不同组分(丹参总酮和总酚酸)对实验性非酒精性脂肪肝(NAFLD)的治疗效果和作用机制。方法:通过喂食高脂饲料复制NAFLD大鼠模型,将大鼠分为正常对照组、模型对照组、东宝肝泰对照组、丹参总酮组和丹参总酚酸组。用药8周后,检测各组大鼠血清中TC、TG、ALT、AST和肝组织中TC、TG的含量或活性变化,用RT-PCR法观察各组大鼠肝脏PPARαmRNA的表达变化,并用HE染色法观察肝组织的形态学变化。结果:丹参两种组分都能降低实验动物血清中TC、TG、ALT、AST的含量或活性,降低和肝组织中TC、TG的含量,促进肝脏PPARαmRNA的表达,改善肝组织病理形态学。结论:丹参总酮和总酚酸防治NAFLD的作用机制可能与增强肝组织中PPARαmRNA的表达,进而促进脂肪酸代谢有关。 Objective:to observe the effect and mechanism of different components of Danshen ( total ketone and total phenolic acid) on non-alcohol fatty liver ( NAFLD) .Methods: Feeding high-fat forage to reproduce NAFLD model rats , divide the rats into control group, model group, Dongbao Gantai group, ketone group and phenolic acid group.8 weeks after administration , detect TC, TG, ALT, AST in blood serum, and TC, TG contents and activity in hepatic tissue;observe the PPARαmRNA expressions with RT-PCR and observe the morphologic variations of hepatic tissue with HE method.Results:both components of Danshen could decrease the con-tents and activities of TC, TG, ATL and AST in serum, decrease TC and TG in hepatic tissue, enhance the PPARαmRNA expression, and improve the pathomorphism of hepatic tissue.Conclusion:The mechanism of Danshen total ketone and total phenolic acid in preven-ting and curing NAFLD may be related to their enhancing PPARαmRNA expression so to improve the metabolism of fatty acids.
