小鼠脾细胞替代人淋巴细胞的E花结试验

SUBSTITUING SPLENIC CELLS OF MICE FORHUMAN LYMPHOCYTES IN THE ROSETTE ASSAY FOR EXPERIMENTAL TEACHING

目的 :探讨以小鼠脾细胞代替人外周血淋巴细胞进行 E玫瑰花结试验实验教学的可行性。方法 :以 0 .41 ml5% SRBC免疫昆明系小鼠 ,4天和 2 1天后制备脾细胞悬液 ,常规方法进行 E玫瑰花结试验。结果 :经 SRBC免疫 4天和 2 1天后 ,与 SRBC混合反应 1 0 min与 1 2 0 min的小鼠脾细胞特异性 E玫瑰花结形成百分率无显著性差异 (P>0 .0 5) ,达 2 8.64± 4.48%至34.38± 7.2 5% ,其形态、大小、染色与人外周血 T细胞 E玫瑰花结相近。结论 :小鼠免疫脾细胞 E玫瑰花结试验可代替人外周血淋巴细胞 E玫瑰花结试验用于实验教学 。

Objective:To investigate the possibility of the E rosette assay in that human lymphocytes could be replaced by splenic cells derived from mice for experimental teaching.Methods:Mice were immunized with 0.4ml of SRBC(5%),prepared the suspension of splenic cells after 4 and 21 days,and then made the E rosette assay.Results:At 4 and 21 day after immunized with SRBC,there was no significance at the percentage of specific E rosette forming cells among the splenic cells mixed with SRBC for 10 or 120minutes(P<0.05).They were from 28.64±4.48% to 34.38±7.25%.The morphology,size and the pattern of staining of mice splenic cells were similar to that of human T lymphocytes.Conclusion:Substituting mice splenic cells for human lymphocytes in the E rosette assay can be used in the students experimental teaching.The result may also show the function of specific humoral and cellular immunity in experimental animals.
