摘要
目的:优选大孔树脂纯化复方酪萨维口腔速崩片中蔷薇红景天提取物的工艺条件。方法:以红景天苷、酪萨维的含量及回收率为指标,通过单因素试验筛选大孔树脂型号并优选其纯化工艺。结果:AB-8型树脂分离效果较好,最佳工艺为蔷薇红景天提取物加适量水充分溶解以2 BV·h-1的流速上样,径高比1∶7,红景天苷、酪萨维的比上柱量分别为7.18,10.43mg·g-1,水洗除杂,加40%乙醇12 BV洗脱,洗脱流速4 BV·h-1,树脂可重复利用3次;红景天苷、酪萨维平均提取量分别为1.870%,3.186%,平均回收率分别为82%,96.26%,平均纯度依次为5.72%,9.72%,纯度分别提高了2.51,2.94倍,浸膏得率由纯化前的13.73%减少至4.57%。结论:AB-8型大孔树脂对红景天苷和酪萨维具有良好的纯化效果,优选的工艺稳定可行。
Objective: To optimize separation and purification technology of extract of Rhodiola rosea from compound rosavin rapidly disintegrating oral tablets by macroporous resin. Method: Taking contents and recoveries of salidroside and rosavin as indexes, single factor tests were adopted to screen macroporous resin model and optimize its purification process. Result: AB-8 macroporous resin had the best separation efficiency, optimum purification process conditions were as follows: added just the right amount of water to ample dissolution of extract of R. rosea, absorption velocity 2 BV .h^-1, diameter-height ratio 1:7, adsorption capacity of salidroside and rosavin 7.18, 10.43 mg .g^-1, washed impurities with water, eluted by 12 BV of 40% ethanol, elution flow rate 4 BV .h^-1, regenerated resin after using three times; Average extraction amounts of salidroside and rosavin were 1. 870% and 3. 186% , average recoveries were 82% and 96.26% , average purities were 5.72% and 9.72% , respectively; Extract yield reduced to 4.57% from 13.73%. Conclusion: AB-8 macroporous resin had a good purification effect of salidroside and rosavin, optimized purification process was stable and feasible.
