三七花多糖的分离纯化及结构初步研究 被引量:13

摘要 热水回流法提取三七花中的多糖,脱蛋白后经DEAE-Sepharose Fast Flow柱层析纯化,得到三部分组分:PNF I、PNF II、PNF III,高效凝胶渗透色谱法进行纯度分析,得到PNF II为一对称峰形。PNF II的重均分子量和数均分子量分别为8.44×104Da和2.47×104Da。气相色谱法分析了精制三七花多糖、PNF I、PNF II、PNF III的单糖组成及其摩尔比,其组成单糖均含有阿拉伯糖、半乳糖、葡萄糖、鼠李糖、木糖和甘露糖,其中PNF II的主要组成单糖为阿拉伯糖和半乳糖。红外光谱和核磁共振分析显示PNF II含有糖醛酸。 The crude polysaccharide was extracted from Panax notoginseng flower(PNF) by hot-water and deproteinized by a method of combining Trichloroacefic acid method and sevage method. Three fraction of this polysaecharide PNF l, PNF II and PNF III were purified through DEAE-Sepharose Fast Flow column. PNF II was identified by high-perform- ance gel permeation chromatography to be homogeneous. The average molecular weight (Mw) and the numerical molecu- lar weight (Mn) of PNF II were 8.44 x 104 Da and 2.47 x 104 Da respectively. The refined polysaccharide, PNF I, PNF II and PNF III were hydrolyzed by trifluoroacetate, and then converted to alditol acetates derivatives, analyzed monosac- charide composition and mole ratio by GC. The results showed that the monosaceharide composition of the refined poly- sacehafide, PNF I, PNF II and PNF III were arabinose, galaetose, glucose, rhamnose, xylose and mannose. PNF II was mainly composed of arabinose and galactose. The IR and NMR spectrum analysis indicated that PNF II contains uronic acid.
