摘要 本文探讨了地龙蛋白肽的氨基酸组成及对血管紧张素转化酶(ACE)活力的影响。采用中医中药的方法将地龙匀浆、浸泡于60℃水浴中1 h,提取活性成分;通过紫外分光光度法测定了地龙中蛋白质的含量;采用凝胶过滤层析法提纯地龙中的活性蛋白并进行体外ACE抑制活性试验,收集活性峰行SDS-PAGE电泳及高效液相色谱法(HPLC)测定地龙蛋白肽的相对分子量及氨基酸的组成。结果表明,地龙蛋白肽含有18种氨基酸,其中亮氨酸和谷氨酸的含量最高,同时含有人体必需的8种游离氨基酸。提示中药地龙中含有抑制ACE的活性物质。本研究为从中药地龙中开发防治高血压病的药物或保健品提供了可靠的实验依据。 This paper aimed to explore the component analysis and the impact of the angiotensin-converting enzyme ac- tivity of earthworm peptide. The active ingredients were extracted from the dried earthworm which was homogenated and soaked in 60 ℃ water bath for 1 h. The protein contents of earthworm were detected by UV speotrophotometry, purified and collected by gel filtration(GF) chromatography. ACE inhibitory activity test was given for the above protein product in vitro and the active peak was collected. The relative molecular weight and the amino acid components of collected pro- tein were detected by SDS-PAGE electrophoresis and high performance liquid chromatography ( HPLC ). The results showed that the earthworm peptide eontains 18 amino acids ,leueine and glutamic of the highest,included eight kinds of free amino acids essential. These findings suggested there were inhibitory ACE activity protein in earthworm, whieh might provide experimental support for further exploration and utilization of earthworm. This study provided a reliable experi- mental basis for developing the prevention and treatment of hypertension drugs or health eare products in the near future.
