摘要
采用超声波协同碱性蛋白酶、木瓜蛋白酶的生物法来水解剩余污泥,以蛋白质提取率为指标,分析了在超声预处理的条件下,这两种不同酶对剩余污泥处理效果的影响。结果表明,碱性蛋白酶水解提取法的最佳条件是:碱性蛋白酶的浓度为6%,固液比为1∶6,水解温度为50℃,水解时间为4 h;蛋白质的提取率为65.72%。木瓜蛋白酶水解的最佳条件是:木瓜蛋白酶的浓度为6%,固液比为1∶4,水解温度为50℃,水解时间为5 h;蛋白质的提取率为62.63%。在优化的试验条件下,超声波协同碱性蛋白酶提取剩余污泥中蛋白质的效果要优于协同木瓜蛋白酶。
Excess sludge was hydrolyzed by combination of ultrasonic and alkaline protease or papain. The influence of two different enzymes on sludge treatment effect was investigated by protein extraction under ultrasonic pretreatment. The results showed that the optimum conditions of alkaline protease hydrolysis were: protease content 6% , the ratio of solid to liquid 1:6, temperature 50℃ and reaction time 4 h. Under these conditions, the protein extraction rate was 65.72%. The optimum conditions of papain hydrolysis were : papain content 6% , the ratio of solid to liquid 1:4, temperature 50℃ and reaction time 5 h. The protein extraction rate here was 62.63%. The combination of ultrasonic and alkaline protease had an advantage over uhrasonic-papain for extracting protein from excess sludge.
