摘要
目的探讨和监测单采血小板保存期间代谢指标的变化,为确立单采血小板体外保存时限提供实验依据。方法应用全自动血气分析仪分别检测保存4h、1d、2d、3d、5d、7d、8d的30份单采血小板的pH值、PO2、PCO2、葡萄糖和乳酸的含量变化。结果单采血小板体外保存8d,葡萄糖持续消耗,乳酸逐渐堆积;pH值在4h至1d短暂上升后持续下降,但最小值大于6.4;P02保存8d最小值为39.2mmHg,远大于2mmHg。结论血小板体外保存8d后,pH值显示血小板仍可能保持其正常功能;PCO2和PO2可不作为评价血小板保存质量指标,适量增加血小板保存液中葡萄糖的含量有利于延长血小板保存时间。
Objective To provide experimental basis of maximum storage of apheresis platelets in vitro through dynamic observation of metabolic changes. Methods Thirty samples were detected for pH,POz ,PCO2 ,glucose and lactic acid content by automatic blood gas analyzer during the storage for 4 h,1 d,2 d,3 d,5 d,7 d and 8 d. Results When platelet preserved to the 8th days in vitro glucose consumption continued and lactic acid was accumu- lated~ pH value declined after a temporary rising in 4 h--1 d,the minimum pHi6.4~ the PO2 showed minimum 39. 2 mmHg which was far higher than 2 mmHg. Conclusion When platelet stored to 8 day in vitro,pH value was still suitable for maintaining platelets' normal function; evaluation of platelets stored metabolites can be ruled out PCO2 and PO2 of these two indicators; modest increase of glucose content in platelet preservation fluid may conducive to the extension of platelet storage time.
