海南黑山羊MSTN基因的表达差异与发育性变化研究 被引量:12

摘要 采用SYBR Green实时荧光定量PCR技术检测了MSTN基因在2、3、4月龄和12月龄海南黑山羊腿肌、腰肌和背肌组织中mRNA表达发育性变换规律,为揭示MSTN基因的生物学功能及其对肉质的表达调控机理提供参考依据。结果表明,在海南黑山羊的四个生长阶段,MSTN基因均在背肌里的表达量最高,腿肌里的表达量较低,表达模式为:背肌>腰肌>腿肌;腿肌和腰肌在2月龄时,表达量较低,3月龄时极显著升高(P<0.01),4月龄时又极显著下降(P<0.01),12月龄时又极显著升高(P<0.01);表达模式为:12月龄>3月龄>4月龄>2月龄;背肌在2月龄时表达量也较低,3月龄时极显著升高(P<0.01),达到最高水平,4月龄时又极显著下降(P<0.01),12月龄时有所上升,但差异不显著(P>0.05),表达模式为:3月龄>12月龄>4月龄>2月龄。 The mRNA expression differences and developmental changes of Calpastatin (MSTN) gene in the leg,waist and back muscle of Hainan Black Goat from 2, 3, 4 and 12 months were investigated by SYBR Green real-time PCR technique, which provides a reference for exploring the biological function of MSTN gene and its expression and regulation mechanism of meat. The results showed that, in the four growth stages, the MSTN gene mRNA expression level in back muscle was the highest and that in leg muscle was the lowest, the expression pattern was: back muscle ) waist muscle 〉 leg muscle, the expression level of leg muscle and waist muscle was very low at month 2, then, it significantly increased (P d0.01) at month 3 and significantly decreased (P^0.01) at month 4, then it significantly increased at month 12 (P%0.01) ; the expression pattern was: month 12)month 3)month 4)month 2. The expression level of back muscle was lower at month 2, but it reached the highest level at month 3 (P〈0.01), then it decreased significantly at month 4 (P〈0.01) and increased at month 12, but it was not significant (P)0.05), the expression pattern was: month 3〉 month 12〉 month 4〉 month 2.
