干细胞标志物Nanog基因检测在肺癌诊断中的临床意义 被引量:2

摘要 目的观察干细胞标志物Nanog基因在肺癌组织与癌旁组织中的表达情况,明确其在肺癌诊断中的作用。方法选择2011年1月至2013年7月在我院诊治且临床被诊断为肺癌的患者作为研究对象,分为肺腺癌(A组)和肺鳞癌(B组)。取患者癌组织与癌旁组织,以实时定量RT-PCR反应检测两组患者干细胞标志物Nanog基因的表达水平,并比较不同分化程度下各组织中Nanog基因的表达量。结果肺腺癌与肺鳞癌患者各年龄段的发病率大致相等,两组患者在性别、疾病分期及有无淋巴结转移方面无显著差异(P>0.05);与I期、II期相比,Ⅲ期、Ⅳ期癌组织中Nanog基因的表达明显升高,且癌组织中Nanog基因的整体表达明显高于癌旁组织,差异有统计学意义(P<0.05)。与中分化及高分化型的癌组织相比,未分化、低分化癌组织中Nanog基因的表达明显升高,差异显著(P<0.05)。结论肺腺癌与肺鳞癌患者癌组织中Nanog基因的表达明显增强,以Ⅲ期、Ⅳ期肺癌中表达最高,细胞的分化程度越低,其癌组织中Nanog基因的表达越强。 Objective To observe the expression of stem cell markers Nanog gene in lung cancer and adjacent tis- sues,specifically its role in the diagnosis of lung cancer. Methods Lung cancer patients were choosed as research sub- jects from January 2011 to July 2013 in our hospital,divided into lung adenocarcinoma (Group A) and squamous cell carcinoma (Group B ) in patients, taking cancer tissues and Adjacent tissues, Measured gene expression by real-time quantitative RT-PCR reactions in patients with stem cell markers Nanog,Contrasted Nanog gene expression in various tissues under different degree of differentiation. Results The incidence rate of roughly equal with squamous cell lung cancer patients of all ages, the two groups in gender, disease stage and lymph node metastasis was no significant differ- ence(P〈0.05) ; Compared with I , ]I stage carcinoma, Nanog gene expression were significantly increased in stage III , IV stage carcinoma,and the overall expression of Nanog gene in cancer tissues were significantly higher than in adja- cent tissues,the difference was statistically significant(P〈0.05). Compared with moderately differentiated and well- differentiated cancer tissue,Nanog gene expression was significantly increased in undifferentiated, poorly differentiated carcinoma,significant difference(P〈 0.05). Conclusion Nanog gene expression is significantly increased in patients with lung adenocarcinoma and squamous cell carcinoma, III, IV stage lung cancer have higher Nanog gene expression, the lower the degree of cell differentiation,the stronger Nanog gene expression in cancer tissue.
