摘要
目的采用蛋白质组学方法,筛选儿童哮喘诊断相关的蛋白靶标。方法使用二维差异凝胶电泳技术(2D-DIGE),分离及筛选稳定期哮喘儿童48例及12例健康儿童血浆蛋白,筛选蛋白进一步经基质辅助激光解吸/电离-飞行时间质谱(MALDITOF/TOF MS)进行鉴定。组间比较采用独立样本t检验。结果通过蛋白质组学分析,组间共筛选出32个差异蛋白(P<0.05),13个蛋白点经质谱鉴定,代表了5种已知蛋白及2种未知蛋白,分别为抗凝血酶Ⅲ(AT-Ⅲ)、α2巨球蛋白(A2M)、补体C3、补体C4a及IGKGIgkappa。其中AT-Ⅲ在哮喘组表达高于对照组(P<0.05);补体C3、A2M在对照组高于哮喘组(P<0.05)。结论通过蛋白质组学方法发现了儿童哮喘病情相关系列蛋白靶标,AT-Ⅲ、A2M、补体C3可能作为未来辅助诊断儿童哮喘的系列蛋白靶标。
Objective using proteomic method upon plasma of stable asthmatic children to find asthma related bio- markers. Methods two dimensional differential gel electrophoresis was used to separate and screen plasma samples of stable asthmatic children and healthy controls for differential expression of proteins. The candidate proteins were identi- fied by Matrix-Assisted Laser Desorption/ Ionization Time of Flight Mass Spectrometry. Results by proteomic tech- niques,32 proteins were found differentially expressed between asthmatic groups and healthy control (P〈0.05), 13 proteins spots were identified by Mass Spectrometry which represent 5 known proteins and 2 unknown proteins. The 5 known proteins were antithrombin- III ( AT- III ), Complement3 (C3), Complement3 a (C3 a), alpha2-macroglobulin ( alpha2- MG) and IGKGIgkappa. AT-III, C3, alpha2-MG differential expressed between asthmatic group and healthy control group. In the asthmatic group the expression levels of AT-III were higer than the healthy control group(P〈0.05) ,the expression levels of C3,A2M were lower than the control groups(P%0.05). Conclusion proteomics is useful in identi- fy asthma related novel biomarkers. AT-III, alpha 2-MG,C3 relate with childhood asthma. The combination of the 3 bio- markers is powerful in evaluating and diagnoseing childhood asthma.
