摘要
目的观察泽泻汤总提物、100%和50%乙醇部位对高血症大鼠血脂代谢及血脂代谢相关酶的影响及差异,在此基础上探寻其可能的作用机制。方法雄性Wistar大鼠喂饲大鼠高脂饲料4周,测定空腹血脂水平,确认高血症模型成功建立后,将其随机分为模型组、泽泻汤总提组、100%乙醇部位组、50%乙醇部位组,另设正常组。连续给药4周,给药期间,正常组给普通饲料,其余各组均给高脂饲料。测定大鼠血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)及肝组织中血脂代谢相关酶羟甲基戊二酰辅酶A(HMGCoA)、低密度脂蛋白受体(LDL-R)、7α-羟化酶(CY7PA1)的mRNA表达。结果泽泻汤总提物及100%和50%乙醇部位组均能显著降低高血脂症模型大鼠血清中TC、TG和LDL-C的含量,明显升高血清中HDL-C含量;可逆转肝组织中升高的HMG-CoA和ApoA-ⅠmRNA表达水平和降低的LDL-R和CY7PA1 mRNA表达水平,其中以泽泻汤总提物和100%乙醇部位为最好。结论泽泻汤总提物及100%和50%乙醇部位均具有降血脂作用,其作用效果以泽泻汤总提物和100%乙醇部位为最佳,调节TC的生物合成和代谢转化,可能是其降血脂的重要机制之一。
Objective Serum total cholesterol concentration is reduced by giving Traditional Chinese Medicine, but the mechanism of Traditional Chinese Medicine on blood cholesterol are uncertain. Methods Hyperlipidemic rats were given distilled water, different Tradi- tional Chinese Medicine (TCM) compounds for 4 weeks. Then Total cholesterol, HDL- C and TG concentrations in the serum and relative quantities of mRNAs in the liver were determined. Results The body weight, food intake, liver weight, and urine volume were not signifi- cantly influenced by TCM. The serum total cholesterol (TC), triglyceride (TG), and serum low density lipoprotein cholesterol (LDL- C) concentration in the CP group were significantly higher than that in the TCM groups, whereas the high - density lipoprotein cholesterol ( HDL - C) concentration in the CP group was significantly lower than that in the other groups at the end of the experiment. The hepatic LDL recep- tor mRNA level in the TCM1 and TCM2 group was significantly higher than that in the CP group, whereas the hepatic LDL receptor mRNA level in the other TCM groups were not significantly influenced compared with that in tKe CP group. The hepatic 3 - hydroxy - 3 - methyl - glutaryl -CoA reductase (HMG -CoA reductase) mRNA level in the TCM1 ,TCM2 ,TCM4 and TCM5 group was significantly lower than thatin the CP group. The cholesterol 7ct - hydroxylase ( CYPTA1 ) mRNA level in the TCM groups were significantly higher than that in the CP group, and The hepatic apo A -I mRNA level in the TCM2 and TCM4 group was significantly lower than that in the CP group. Conclusions These findings suggest that the cholesterol lowering effect of TCM in hypercholesteremic rats is Caused at least in part by increased hepatic LDL - R and CYP7a - 1 expression and decreased HMG - CoA and APO A - I expression.
