摘要
目的 :观察新的二氢吡啶类钙拮抗剂 MC92 0 4对大鼠肾性高血压的治疗作用。方法 :采用一肾一包扎型肾性高血压模型 ,测定不同条件下动物血压、心脏重量、肾血流量 (RBF)和肾小球滤过率 (GFR)。结果 :术后 4周开始给予 MC92 0 4(2 0 m g· kg- 1 ,ip,b.i.d) ,可显著降低心脏湿重与体重比和左心湿重与体重比 (P<0 .0 1) ,改善心肌肥厚症状。静脉连续输入 MC92 0 4和 nifedipine(Nif) ,在 1~ 10 μg· kg- 1· min- 1范围内 ,有显著的降压作用 ,并表现出剂量依赖性 ,MC92 0 4降压效应明显大于等剂量 Nif。同时发现 ,MC92 0 4和 Nif在 1μg· kg- 1· min- 1时 ,均可显著增加 RBF和 GFR(P<0 .0 5 ) ,且 MC92 0 4增加 RBF的程度比 Nif显著。但这一效应在 3,10 μg· kg- 1·min- 1 时 ,由于血压过低而逆转。结论 :MC92 0 4对大鼠肾性高血压具有较好的治疗作用 。
AIM: To study the therapeutic effects of a novel dihydropyridine calcium antagonist,MC9204, on renal hypertension of rats. METHODS: Using one kidney one clip renal hypertensive model, we determined the arterial pressure, heart weight, renal blood flow(RBF) and glomerular filtration rate(GFR) at varied treatment. RESULTS: Given from 4 weeks after operation,MC9204 (20 mg·kg -1 ,ip, b.i.d) significantly decreased the ratios of heart wet weight to body weight(HWW/BW)and left heart wet weight to body weight(LVWW/BW)( P <0.01),reversing myocardial hypertrophy. At the dose of 1~10 μg·kg -1 ·min -1 MC9204 and nifedipine(Nif) induced hypotensive reaction in the dose dependent manner, and Mc9204 showed more potential hypotensive reaction than that of Nif at the same dose. In the meantime MC9204 and Nif notably increased RBF and GFR at 1 μg·kg -1 ·min -1 ( P <0.05), while MC9204 showing stronger than Nif. However, this effect was reversed at 3 and 10 μg·kg -1 ·min -1 for the excessive hypotension. CONCLUSION: MC9204 had satisfying therapeutic effects on renal hypertension of rats, and showed bright prospects in clinic.
