生、煨葛根对大鼠离体十二指肠平滑肌运动的影响 被引量:5

摘要 目的观察生、煨葛根对大鼠离体十二指肠平滑肌运动的影响,以探讨葛根炮制前后的止泻作用差异和相关作用机制。方法采用离体肠张力检测装置,以平均张力为主要指标,观察生、煨葛根对大鼠离体十二指肠平滑肌自发运动的影响以及氯化钡(BaCl2)、乙酰胆碱(Ach)、去甲肾上腺素(NA)、异丙肾上腺素(ISO)与生、煨葛根对肠肌作用的相互影响。结果生、煨葛根在2.4~29.1g/L范围内能抑制肠肌的自发运动,使平均张力降低且呈剂量依赖性;生、煨葛根低(5.0g/L)、中(14.8g/L)、高(29.1g/L)剂量均能抑制BaCl2引起的痉挛性收缩;生、煨葛根对Ach引起的肠肌兴奋有拈抗作用,但Ach对生、煨葛根抑制作用后的肠肌未显示出兴奋作用;生、煨葛根对NA、ISO引起的肠肌抑制有抑制作用,NA、ISO对生、煨葛根引起的肠肌抑制仍能表现出抑制作用。结论生、煨葛根均能抑制大鼠离体十二指肠平滑肌运动,煨葛根较生葛根作用明显;其作用机制可能是通过阻断M受体或直接作用于平滑肌,降低平滑肌细胞内Ca2+浓度实现的。 Objective To observe the effects of raw and bran-roasted Radix Puerariae on movement of duodenal smooth muscle of rats in vitro, and to discuss the different in antidiarrheal effect of Radix Puerariae before and after processing and its mechanism of action. Methods Using intestinal detection device in vitro and taking average tension as the leading indicator, the inhibitory, effects of raw and bran-roasted Radix Puerariae on rat smooth muscle of duodenum contractility in vitro and its interreaetion with BaC12, Aeetylcholine (Aeh) , Noradrenaline bitartrate (NA), Isoprenaline Hydroehlofide (ISO) were observed. Results Raw and bran-roasted Radix Puerariae could inhibit rat smooth muscle of duodenum spontaneous contraetions in 2.4-29.1 g/L, reducing the tone of contractions dose- depcmtently; raw and bran-roastcd Radix Puerariae of 5.0 g/L, 14.8 g/L, 29.1 g/L could suppress the spasmodic contraction induced by BaC12 ; both raw and bran-roasted Radix Puerariae could against myentcron stimulation induced by Ach, but no excitatory effect was found in Ach on smooth muscle of duodenum pretreatment with raw and bran-roasted Radix Puerafiae; both raw and bran-roasted Radix Puerafiae had inhibitory effect on intestinal inhibition induced by NA and ISO, aud NA, ISO showed inhibitory effect on intestinal inhibition induced by raw and bran-roasted Radix Puerariae. Conclusion Both raw and bran-roasted Radix Puerariae have inhibitory effect on the movement of duodenal smooth muscle in rats in vitro, and the inhibitory effect of the later is more obviously. Its mechanism may be realized by blocking M-receptor or acting on smooth muscle, and reducing the Ca2+ concentration.
